ELISA- Enzyme-Linked Immunosorbent Assay

ELISA- Enzyme-Linked Immunosorbent Assay (also called “enzyme immunoassay,” or EIA) is a microtitre plate based quantitative technique in which an antibody is linked to an enzyme (preferably HRP or AP) prior to reaction with the antigen and is then followed by assessment of the enzyme conjugated antibody–antigen reaction upon incubation with the enzyme-specific substrate. The reaction produces a detectable signal as change in the substrate color that could measured at specific wavelength.

There are four major types of ELISA

  • Direct ELISA
  • Indirect ELISA
  • Sandwich ELISA
  • Competitive ELISA

Most of plant pathogen diagnostic ELISA kits designed to be highly sensitive and specific. So, it use more than one antibody system which falls under Sandwich ELISA.

There are two types of sandwich ELISA

1. Double antibody sandwich ELISA (DAS- ELISA): In this assay, monoclonal capture antibody (CAB) and polyclonal detection antibody with enzyme conjugate sandwiches the antigen of interest. This is then incubated with substrate that reacts with the enzyme and develop colour. This protocol allows sensitive and specific detection of pathogen/ antigen of interest.

2. Triple antibody sandwich ELISA (TAS- ELISA)/ Compound-ELISA: In this assay, capture antibody (CAB) and detection antibody sandwiches the antigen of interest. This is further incubated with antibody – enzyme conjugate. The enzyme reacts with substrate and develop colour which could be read visually or in ELISA plate reader.

ImmunoStrip

ImmunoStrip®:  The rapid test/ Lateral flow immunostrips- ISK/STX are available for more than 24 pathogens can easily test in the field, with lab-quality results in 30 minutes. Get the answers you need, when you need them!

  • Select and Extract Sample:  Select sample from plant and insert into the mesh-lined bag. Grind by rubbing the outside of the bag with a blunt object.
  • Insert ImmunoStrip:  Insert ImmunoStrip Into the extraction bag with the green end facing down. Allow the test to run 30 minutes.
  • Interpret Results: If the sample is negative, only the control line will be visible. If the sample is positive, a colored test line will appear below the control line.

ImmunoComb®:  These tests are a rapid means of screening crops for the presence of multiple pathogens. ImmunoCombs consist of four ImmunoStrips attached together for ease of testing. ImmunoComb tests require no equipment or expertise to run. Results are obtained in as little as a few minutes making them perfect for use in the field or greenhouse.

AmplifyRP® Isothermal Amplification

Agdia AmplifyRP® comes in two different formats- Acceler8 and XRT.

AmplifyRP® Acceler8® : AmplifyRP® Acceler8® is a rapid RNA amplification and detection platform designed for field-based or laboratory testing of different plant pathogens. It is formulated to allow an end-point detection of amplified products using a lateral flow strip. This lateral flow strip is housed inside an Amplicon Detection Chamber that is specifically designed to prevent the escape of amplified products and thereby also prevent laboratory contamination. AmplifyRP® Acceler8®enables users to rapidly amplify RNA at a single operating temperature using only a crude sample extract. Amplified product is then tested in an Amplicon Detection Chamber where clear "yes / no" results are read visually on a lateral flow strip (much like a pregnancy test). The entire testing process can be completed in as little as 30 minutes.

AmplifyRP® XRT and XRT+

XRT and XRT+ kits provide real-time isothermal nucleic acid amplification and detection in a portable and easy-to-use format. XRT reaction pellets contain Exonuclease III which will process suitably designed probes during the amplification step and produce a real-time read out much like a real-time PCR. Using AmplifyRP, end-users can obtain the same high level of sensitivity and specificity as PCR but with some distinct advantages.
